splitting and moving a hive

I am planning to split a hive this weekend.  I am purchasing a new queen to put into the hive.  The split will actually be a full 10 frame box of already established bees.  That's because I added a deep box onto the lower box a few weeks ago to help prevent a swarm (which seemed to work) even though here in FL I only need one deep per hive.  So I plan to take the top deep and move it to a location about 45 min away.  Question:  Should I move the box one day then go back and introduce the new queen the next day?  If so should I let the workers have immediate access to the sugar plug or let her stay in the cage a few extra days?  And does it matter what time of the day I move the box (evening, to get as many bees as possible?)

I would think it is better to move in the evening because most of the bees are home and it is cooler.  They say you can requeen after about 4 hours, but I have always waited about 24 hours and have had good success.  I was told not to remove the candy plug for about 2 days, and then to do so only if they appear receptive to the new queen.  This is the way I have done it, but I also didn't have to travel 45 minutes each visit.  There may be shortcuts to this process, but I still would take the cautious approach because queens are expensive and it is time consuming to mess around with the whole process, so you might as well do it right and have it take.

Just plan what you want to do.  If you want to minimize drifting or you want to maximize it.  Moving after dark will have the bees home and some will reorient.  Moving in the afternoon will have most of them gone who will, of course, fly right back to the old location.
